Studies on Effects of Factors on F1 Anther Culture from Japonica/Indica Hybrid Rice

Abstract: The anther of Jindao291/IRBB60 and their parents were used to induce the calli in the different medium supplemented with different concentrations and combinations of phytohormones and organic nutriments and different high temperature pre-culture treatments. The results showed that the frequency of the callus induction in the SK3 medium supplemented with 2 mg/L 2, 4-D, 1 mg/L NAA and 1 mg/L KT was 0.67%-2.78% higher than that with 2 mg/L 2, 4-D. The induction rate could be increased in this medium added with hydrolyzed casein. It could be obtained the highest plant regeneration rate in the MS medium added with NAA (1 mg/L), 6-BA (2 mg/L) and KT (0.5 mg/L). The frequency of the callus induction and green plantlet differentiation and anther culture ability of the cross F1 is significantly higher than that reverse F1. Anther culture ability may be the highest when anthers were pre-cultured with 30℃ for 36 h.
