Analysis of Quality Characteristics of Wheat Double Haploid Lines Generated Via Anther Culture

Abstract: In this paper, we systematically tested the quality characteritics of wheat double haploid(DH)lines from the first generation(H1) to H5, with the aim to breed elite winter wheat lines with superior grain quality trait. We found that the composition of high molecular weight glutenin subunits did not change from H1 to H5. By contrast, seed protein content varied significantly between H1 to H2. Additional analysis revealed the existence of high conelation betweenthe test results of H2 and H3 generations with respect to seed protein content, grain hardness and SDS sedimentation volume of dough. Through early tests of quality characteristics, a DH line(Jingdan 00-594) shoving superior grain quality trait was developed. Its grain and dough properties benene stable in as early as H4 and H5 genenations. Based on our data, an effective protocol for the early and efficient selection of the DH lines with elite grain quality trait is proposed and discussed.
