Production of Agrobacterium-mediated Transgenic Potato Plants Containing phaG Gene

Abstract: phaG is a key enzyme in PHAS biosynthesis process.In order to produce medium-chain-length polyhydroxyalkanoates(mcl-PHAS)in transgenic plants,plant expression vector pTiBHS-2 containing phaG cloned from Burkholderia caryophylli(BcphaG) was constructed.A major commercial cultivar of potato,Atlantic,was transformed via Agrobacterium tumefaciens mediated procedure.Nine transgenic plants are confirmed positive by PCR analysis.Western blot analysis demonstrated that BcphaG gene is expressed.The effect of over-expressing BcphaG gene on plant growth and development were also analyzed.At the early stage,compared to control plant,the growth of over-expressing lines is inhibited,root regeneration is delayed.However,these transgenic plants recover gradually after roots regenerated.It suggested that plant growth and development is partially affected by over-expressing BcphaG gene.
