Construction of mtDNA Library of CMS Maintainer of Common Wheat

Abstract: Cytoplasmic male sterility(CMS)line 75-3369A's maintainer 75-3369B of common wheat was studied. mtDNA was digested completely with EcoRⅠ and partly with MboⅠ and two gene libraries(ME and MM)of mitochondrial DNA(mtDNA)of 75-3369B was constructed. ME consisted of 350 clones and MM contained 450 clones. MM library included 98.12 percent of the mitochondrial genome. The advantage and disadvantage of the two methods of gene library construction were also discussed.
