A PCR-Based Diversity Analysis of Waxy Genes in Shaanxi Wheat Cultivars and Lines

Abstract: Three optimized STS markers were used to analyze 99 wheat cultivars in Shaanxi from 1960 with different types of Waxy proteins. The results indicated that the marker on Wx-A1 locus amplified 336 bp and 317 bp special fragments associated to Wx-A1aand Wx-A1b genotypes; the marker on Wx-B1 amplified 425 bp special fragment associated to Wx-B1a and null special fragment associated to Wx-A1b genotype and the marker on Wx-D1 amplified 867 bp and 279 bp special fragments associated to Wx-D1aand Wx-D1bgenotypes, respectively. Except for the cultivar Shaannuo 1 with the genotype of Wx-A1b + Wx-B1b + Wx-D1b,12 cultivars were identified to null Wx-B1 mutants and no cultivars were detected for null Wx-A1 mutants and null Wx-D1 mutants. The three optimized STS markers were demonstrated to be useful tools to identify wheat cultivars with mutant and normal alleles of the Waxy genes in marker-assisted selection of wheat breeding programs and the genotypes of Waxy genes of some materials were released in this paper.
