Establishment of the Transfer System by Agrobacterium-mediated and Transformation of Foreign gene' for Capsicum frutescens

Abstract: The transfer system by Agrobactericum-mediated for Capsicum f rutescens included six steps:preparation of sterilize plant, culture of bacteriaLstrain, transformation by Agrobacterium-mediated, shoot induc-tion, shoot elongation, root induction and transplanting.A better shoot induction medium was MS+4-6mg/L6-BA+0.5mg/LNAA+2.0mg/LAgNO3, the frequency of shoot regeneration was different with various breeds, and the highest frequency reached more than 70%.The cotyledons frequency of shoot regeneration was obviously higher than the hypocotys.A better shoot elongation medium was MS+3.0mg/L6-BA+0.3mg/LNAA+1.0mg/LGA3and the elongation ratio was 40%.T he medium 1/2 MS+0.3mg/LNAA(or 0.5mg/LIAA) was much suitable for root induction and the average ratio may reach 65%.Plants with insect-resistant gene had been obtained through the study on three Capsicum f rutescens breeds with the method of Agrobacteri-um-mediated.
