Study on the Transformation of Cotton Mediated with Agrobacterium tumefacines

Abstract: Hypocotyl used as explants,twenty varieties or lines were studied for plant regeneration or somatic embryoid.Nineteen of them were successful.According to the result,the varieties easy in plant regeneration Han 93-2,Hanwu 2031,Hengwu 89-30,Coker 201,Coker 312 were chosen for the transformation.Inoculating rate of Kanamycine resistant callus was about 50%,among them 40% was GUS positive.50% of plants regenerated from the GUS positive callus were GUS positive,and proved by Southern hybridization analysis.
