Studies on Gene Transformation by Introducing Exogenous DNA into Cucumber After Pollination

Abstract: Cucumber (Cucumis sativus L.) cultivars Jinyansihao being chosen as the recipient and the plasmid pBI121 being chosen as donors,the research of the technique of direct exogenous DNA introduction after artificial pollinationhave been conducted.The genetic transformation system of cucumber withhigh efficiency by injecting exogenous DNA into ovary after pollination were established,and transgenic plants were obtained.Studies showed that the fruit setting ratio and the number of seed per fruit were the lowest by the method of ovary injection,while thehighest transformation ratio were got by this approach.The injection of DNA solution at about 12hours after pollination resulted in thehighest transformation rate,while the concentration、pH value and the volume of DNA solution were important factors affecting fruit setting and seed production.GUS analysis and southern blottinghybridization results showed that NPTⅡ and GUS gene were transformed into cucumber.
