Callus Induction and Multi-bud Body Formation from Stem Segment of Dioscorea opposita

Abstract: The studies on in vitro culture of stem segments with bud and budless segments of Dioscorea opposita showed: (1) In the same culture condition, the budless stem segments were more advantageous to callus formation, but different phytohormones and their combinationhad different effects on callus induction and the most suitable combination were MS+6-BA 1 mg/L+NAA 0.5 mg/L. The stem segments with bud were more favourable to multi-bud body. Multi-bud body with 3-5 buds were formed on MS basic medium containing 6-BA 1-2 mg/L+NAA 0.1 mg/L.On this basis, the number of buds in multi-bud body was increased by adding PP333, but reduced by adding GA, to the medium. (2) A large number of plantlets might be reproduced within a short time by way of the formation of multi-bud body by stem segment with bud, thus, providing a useful way for the rapid popularization and planting of excellent varieties of Dioscorea opposita.
