Studies on Transformation of Maize Inbred Line with Protein Kinase Genes by Pollen-tube-pathway

Abstract: Three plant expression vectors harboring protein kinase genes of ZmPti1,ZmPti1-1,or ZmCIPK2 were transferred into maize inbred line Ji 444 separately by pollen-tube-pathway,and 40 glufosinate-tolerant plants were obtained through herbicide screening,then 28 of them were confirmed positive by PCR analysis.The average transformation rate was 1.33%,and 11 transgenic lines were harvesting T1 seeds.Control plants along with T1 transgenic plants were cultivated under drought conditions,and the yield of each plant and 1000-grains weight were analysis,then their drought tolerance coefficient of T1 transgenic lines was calculated,and the elementary results suggested that transgenic plants was more tolerant to drought stress than that of control plants,and protein kinase can improve the drought tolerance of maize.
