Cloning,Sequencing and Structure Prediction of T Cell Receptor α Genes in Pigs

Abstract: In order to study molecular structure and function of T cell receptor,porcine T cell receptor alpha chain gene was cloned from peripheral-blood lymphocyte(PBL) of Gansu hezuo pig at the first time in domesdic.The sequence analysis shows that the ORF of total pTCR-α gene is 819 bp in length,encoding 272 amino acids with a signal peptide composed of 20 amino acids.The protein molecular weight is about 30 kDa,with 18 Strongly Basic(+) Amino Acids,23 Strongly Acidic(-) Amino Acids,87 Hydrophobic Amino Acids and 102 Polar Amino Acids.The homology of nucleotide sequence of porcine TCR-α with the referenced sequence(AB087988) is up to 94.6%.Compared with other porcine genes of TCR-α published in GenBank,the homology is about 54.4%-80.3%.It may be a novel transcript of TCR-α family.In order to research molecular structure and function of T cell receptor,this study also uses bioinformatics to analyze its gene sequences and structural feature of encoding protein.
