Construction of Selectable Marker Excision Vector forahFAD2RNAi and Its Transformation

Abstract: Napin promoter was ligated withahFAD2RNAi construct,which contains inverted repeat fragments of FAD2 gene from peanut.The selectable marker excision vector pNCX-FAD2RNAi(N)was constructed by subcloning theahFAD2RNAi(N)into expression vector PNCX,which included an estrogen-inducible Cre/loxP DNA recombination system for the purpose to obtain selectable marker free transgenic lines of peanut with high oleic acid.It is confirmed by restricted enzyme digests that theconstruct contains the promoter,FAD2RNAi and NCX-FAD2RNAi fragments.Agrobac-terium mediated transformation was conducted and 214 cluster buds were obtained.
