The Orthogonal Optimization of SRAP Amplification System in Cucumber

Abstract: In this study, the optimum SRAP-PCR reaction system in cucumber was established with orthogonal design on four levels of five factors (dNTP, template DNA, primer, Taq DNA polymerase, Loading buffer). In a total volume of 10 μL SRAP-PCR system, the optimum SRAP-PCR reaction system contains 1 μL 10×PCR buffer, 150 μmol/L dNTP, 30 ng template DNA, 0.3 μmol/L primer, 1.5 U Taq DNA polymerase. The optimum volume of Loading buffer is 10 μL.
