Isolation and Identification for a Wild Fungus of Genus Agaricus

Abstract: The aim of this study is to identify an isolated fungus from wild environment. This study established?a method for the identification of the wild fruiting body forming fungi. Method:The mycelium was isolated from the?fruiting body by using PDA medium. Genomic DNA was extracted from not only mycelium and fruiting body then?used as template for the ITS gene PCR amplification. Further,phylogenetic analysis based on the ITS sequence was?conducted to confirm the genetic position of this fungi. Results:About 750 bp fragment was amplified and sequenced. The results showed that ITS sequence from mycelium and fruiting body was uniform. Phylogenetic analysis?indicated that the fungi belonged to the genus Agaricus. GenBank accession number is JN907015. Conclusion:The?results established an identification method for the wild fruiting body forming fungus and laid the foundation of the?application of this fungus.
