Study on NaCl Screening to Obtain Marker-free and High Effective Salt-tolerant Transgenic Rice

Abstract: It is an effective method to use NaCl selecting to generate marker-free salt-tolerant transgenic rice. We used the three japonica rice cultivar(ZhongHua11, Xiushui11, Chunjiang06) mature embryos callus as materials, SKC1 as the target gene to study that on the conditions of screening NaCl, heat shock treatment and liquid coculture of Agrobacterium-mediated gene transfer salt-tolerant rice efficiency. The results showed that heat shock and liquid co-culture of rice could enhance the efficiency of salt tolerance gene transfer. Among them, the heat shock treatment increased the maximum efficiency of gene transfer was 46. 2%, while the liquid co-culture increased the highest efficiency of gene transfer was 60. 7%.
