A Study on Improving Frequency of Isolated Microspore-derived Embryos of Chinese Cabbages

Abstract: Studies on factors for enhancing theembryogenic rates in isolated microspore culture of chinese cabbages were carried out.The results showed that 21 varieties had produced microspore-derived embryos among 24 varieties of chinese cabbage cultured in NLN-13 liquid medium.The production of microspore-derived embryos varied remarkably among these varieties.Embryo yield per bud is from 0.1 to 14.66.Sucrose and hormone concentrations in NLN medium were compared.NLN with 13% sucrose is better in production of microspore-derived embryos.Average embryo yield is higher in 33℃ high temperature treatment from 24 to 48 hour.The B5 medium with 6-BA 0.2mg/L,3%Sucrose and 1% agar is good for microspore-derived embryo to regenerated plants.
