Construction of AFLP Analysis System in Cucumber

Abstract: AFLP (Amplified Fragment Length Polymorphism) analysis system was established with 24 cucumber self-bred lines. Several factors, include the quantity of genome DNA for cutting, the diluted times of pre-amplifications and the number of selective nucleotide in primer, were optimized. With the optimized system, the AFLP fingerprinting of 24 cucumber materials amplified by primer combination P12M12 have been obtained, which had 35 clear bands, and 4 of them were polymorphic.
