Isolation and in vitro Culture of Sheep Preantral Follicles

Abstract

Abstract: Isolation method and sheep age had significant effects on the isolation efficiency of sheep preantral follicles.Per sheep mechnical isolation obtained the same total preantral follicles(89.5) with that of enezmy digestion (103.3)(P0.05) per sheep,but normal follicle rate(31.2%) of mechnical isolation was high than that (18.5%) of enezmy digestion (P 0.05).By mechnical isolation, per preputuberty sheep obtained higher total preantral follicles and normal preantral follicles(125.6,35.1)than that of adult sheep (60.4,16.1)(P0.05), but normal follicle rate did not differ(28.1 % vers 26.8% ).Preantral follicles were cultured in vitro for 6 days, no difference was observed in oocyte growth (34.8 μm vs 29.9 μm) between IGF-I (20 ng/mL) treated follicles and controls.
