Determing Purity of Corn Seeds with Isoelectric Focusing Electrophoresis

Abstract: Esterase isozymes in more than 150 corn inbred lines and 90 corn hybrids were investigated using polycrylamide gel isoelectric focusing electrophoresis. All of the inbred lines and hybrids could be unequivocally distinguished from one another by their zymograms. According to the zymogram,the purity of the specific corn inbred or hybrid could be counted and showed by the percentage.The germination tests were taken at the same time in the field by using the same com inbred and hybrid.The two tests showed the same results. IEF is a very useful quick and accurate method to determine the purity of corn inbred and hybrid and is widely used in corn breeding program and com production.
