Artificial Inoculation of Wheat Yellow Mosaic Virus to Wheat

Abstract: Factors affecting artificial inoculation of WYMV on susceptible wheat variety were analyzed.The result indicated that the highest incidence could be gained by using the dried diseased-leaf or fresh wheat to inoculate wheat cultivated at low temperature.The diseased-leaf should be grounded in phosphate buffer. The effective inoculation method was developed and WYMV in wheat leaves was tested by ELISA and RT-PCR.
