A study on Factors Effecting Isolated Microspore Culture of Broccoli

Abstract: In the study, 3 genotypes of broccoli, i.e.“Galaxy"、“bc-0”、“bc-4”were used to study factors such as osmotic pressure, exo-hormones, the temperature and time of heat treatment and the length of bud or the stage of pollen development which effect on the isolated microspore culture of broccoli.The exo-hormones were not essential for inducing embryogenesis, but 6-BA alone could produce better results than 6-BA plus NAA.The heat treatment at 32.5℃ for 24hr was the most effective for inducing embryogenesis.The most effective length of buds was dependent on plant genotypes.For example, for “bc-4”at the period of buds 4.1-5.0mm in length, most of the pollens belonged to the late-uninucleate and the early-binucleate stages, it produced 37.08 embryos per bud.
