    To further understand the molecular characterization and expression patterns of SlNAC80, we cloned this gene from tomato SlNAC 80 contains an open reading frame of 1 023 bp encoding a protein of 340 amino acids.The molecular weight and isoelectric point of SlNAC80 protein was 38.19 kDa and 5.27, respectively.SlNAC80 contains a typical NAC domain in the N-terminal, which consists of five subdomains (A to E).Quantitative Real-time PCR (qPCR) analysis showed that SlNAC80 was expressed in all tested tomato tissues, with the highest in green fruits and old leaves, and the lowest in red ripe fruits.Promoter sequence analysis indicated that there were many cis-acting regulatory elements involved in response to hormones (ABA, GA, SA and ETH) and abiotic stresses (cold, dehydration and salt stresses), such as ERELEE4, GARE1OSREP1, LTRE1HVBLT49, GT1GMSCAM4, MYB1AT, MYCATERD1, MYCATRD22, WBOXNTERF3, WRKY71OS, and etc.The qPCR analysis also demonstrated that the expression of SlNAC80 was induced by cold, drought, salt, methyl viologen, ABA and ethephon treatments.These results suggest that SlNAC80 may play important roles in regulating abiotic stress responses in tomato.

    Effects of NaCl stress on seed germination,growth and ionic absorption and distribution of tomato seedlings were studied.Results showed that 25-100 mmol/L NaCl didn't decrease seed germination rate,but delayed the date of seed germination.125-225 mmol/L NaCl obviously decreased seed germination rate and no seed germinated during 250-300 mmol/L.25-300 mmol/L NaCl significantly inhibited seedling growth and decreased dry matter accumulation.Na+ was mainly accumulated in stem and root of the seedlings and with lower content in leaves;Cl- was mainly distributed in stem.K+ content in leaves exposed to salinity increased remarkabledly,and absorption of S-(K,Na) in root and transportation of S-(K,Na) in leaves were uniformly promoted obviously.It suggested that higher selectivity of tomato to K+ during ion absorption and transportion made tomato with salt-tolerance.

    Calmodulin is an important Ca2+ receptor protein in plants,which plays an important role in calcium signaling pathway and stress resistance.It is of great significance to study the mechanism of CaM protein in tomato under low temperature stress for cold tolerance breeding of tomato.Tomato calmodulin genes SlCaM3, SlCaM4 and SlCaM5 were cloned from tomato varieties Heinz1706,LA3969,Jifen 2,Jifen 3 and Nongbofenba 15,and their sequences and protein sequences were analyzed by bioinformatics;the cis acting elements in the promoter regions of SlCaM3,SlCaM4 and SlCaM5 were analyzed by plantcare;Quantitative Real-time PCR was used to analyze the expression patterns of SlCaM3,SlCaM4 and SlCaM5 under 15,5 ℃ temperature stress in different tomato varieties,and the specific expression of SlCaM3,SlCaM4 and SlCaM5 in different tissues was analyzed in combination with RNA-seq data.The results showed that the coding sequence of SlCaM3,SlCaM4 and SlCaM5 were 450 bp,and their similarity was 93.63%;the encoded amino acid sequences were identical,belonging to acidic stable hydrophilic protein with typical conservative domain of cam protein.The analysis of cis acting elements showed that the promoter regions of the three genes contained not only the necessary core elements,but also a variety of biotic and abiotic stress response elements,and showed a complementary pattern.The analysis of the expression patterns of different degrees of low temperature stress showed that the expression patterns of SlCaM3,SlCaM4 and SlCaM5 in five tomato materials showed a trend of first decreasing and then increasing at 15 ℃,and the expression of SlCaM5 increased more significantly.At 5 ℃,SlCaM3 and SlCaM4 did not change significantly,but the expression level of SlCaM5 gene increased significantly in the later stage of treatment.The high expression of SlCaM5 gene in the process of tomato resistance to low temperature indicated that it maintained the translation level of cam protein under low temperature and ensured that the function of CaM protein was not affected.The analysis of SlCaM3,SlCaM4 and SlCaM5 specific expression in different tissues of Heinz1706 showed that SlCaM3 and SlCaM4 were highly expressed in meristem,while the expression of SlCaM5 different tissues was not significantly different.

    In order to identify the pathogenic fungi which causing tomato bacterial wilt in Fengnan District of Hebei Province.This study proved the disease as Bipolaris sorokiniana by pathogen isolation,pathogenicity test,morphological observation,and rDNA-ITS gene sequence analyses was caused by Fusarium oxysporum f.sp.radicis-lycopersici(Forl).The result showed that the mortality and morbidity of stem based inoculation were higher than root soaking inoculation.So,the disease with stem based inoculation developed faster.The study provided theoretical basis for the breeding of disease resistance and the screening of the fungicides.

    The colonization and biocontrol effects of plant growth promoting rhizobacteria-Pseudomonads in the rhizosphere of tomatoes were investigated.Pseudomonas CBT1 can colonize the rhizosphere of cucumbers by the methods of seed soaking and root watering.The antimicrobial activity results showed that some isolates suppressed the mycelial growth of B.cinerea and F.oxysporum f.sp. cucumerinum,the highest inhibitory rate was 89.1% and 65.6%,respectively.The efficient isolates were tested and found relative control effects on grey mold and fusarium wilt diseases of cucumber were 44.0%-67.7% and 47.7%-55.4% in pot experiments,respectively.

    It is necessary for us to clone some endogenous promoters with high transcription activity in tomato leaves and construct CRISPR/Cas9 system, which could provide an effective technology for the research of molecular design breeding in tomato. Six different truncated tomato U3 promoters had cloned by two rounds of PCR amplification from tomato variety Zhongshu 4. Six GUS fusion expression vectors were constructed and transformed into tomato leaves by the agroinfiltration. After right sequencing of the transfer PCR products, the necessary element for transcriptional function of six kinds of SlU3 promoter was analyzed using DNAMAN software. The different truncated SlU3-1P,SlU3-3P and SlU3-4P promoters from tomato variety Zhongshu 4 were performed to clone by two rounds of PCR amplification, altogether there were six different promoters. Lengths were 489,318,450,248,457,248 bp respectively. Six GUS fusion expression vectors were constructed and transformed into tomato leaves.The promoter sequence alignment analysis revealed that the tomato U3 promoter, like the Arabidopsis U3 promoter, also contained two more conserved elements, the USE and TATA boxes, and the positions between the two elements were relatively fixed.Transfected tomato leaves by agroinfiltration transformation method. The results showed that the six truncated U3 promoters could drive GUS expression in tomato leaves and tomato leaves could be stained blue and the shorter promoters also had stronger transcription activity as original promoters.Six different tomato U3 promoters had cloned with high-level transcription in tomato leaves, which could provide ideal endogenetic promoters for the construction of CRISPR/Cas9 system for tomato functional genomics research.

    The effect of soil amendments on soil microbial communities,quality and yield of tomato in protected house were investigated for providing a scientific basis for reasonable use of soil amendments.A field experiment was initiated in a planting tomato greenhouse with different kinds of soil amendments(lime nitrogen,fosthiazate,abamectin and substrate).Changes of bacteria,fungi,quality,yield were monitored.Results showed that four different types of soil amendments could significantly increased the number of soil bacteria,fungi,actinomycetes and antagonists,while the numbers of bacteria and fungi in the later growing period of tomato compared with the middle growing period were decreased;the contents of lycopene and Vc reached the highest under substrate treatment for 8.75 mg/100g,78.60 mg/kg,respectively;the soluble sugar content ranged from 2.85% FW to 3.29% FW;compared with other treatments,the sugar acid ratio of CK,substrate and abamectin were 7.57,6.91 and 7.70,respectively,taste the best;under substrate treatment,the yield of tomato was the highest for 139 059.30 kg/ha,which was increased by 21.18% compared with CK.Considering the taste quality and yield,substrate could be used as a preliminary reference for tomatoes production.

    The inhibitory effect of Streptomyces alboflavus TD-1 against Botrytis cinerea and its effect on defensive enzymes activity in vitro antibacteral activity and pot experiment were studied in order to provide reference for biological control of grey mould.The results showed that inhibition rate of fermentation broth of the strain TD-1 on mycelium growth and spore germination of B.cinerea were 98.1% and 98.3% respectively.The control efficiency of TD-1 fermentation broth against B.cinerea in tomato plants was 76.98% in pot experiment.The activity of PAL, PPO, POD and SOD of tomato leaves increased after spraying fermentation broth of the strain TD-1 and spore suspension of B.cinerea.They reached maximum in the fourth day, the third day, the second day and the third day respectively.

    We examined the effects of cinnamic acid on the growth of tomato seedlings,and its alleviation of applied carbonized maize cob was shown.The tomato seedlings were transplanted in the hydroponic system with perlite as substrate,and the biomass,photosynthesis,ultrastructure of root and MDA were investigated.The results showed that cinnamic acid inhibited the biomass,photosynthesis and chlorophyll contents of tomato seedlings.Deformation of the ultrastructures of root was observed and the MDA content was increased by the treatment of high content cinnamic acid.But the inhabitations were alleviated by applied carbonized maize cob.Carbonized corn cob can be used to prevent the plants from monocropping obstacles.

    Fifty-eight tomato genotypes with different lycopene content were used to study the relationship between their lycopene content and color readings of CR400.Experimental results showed that L1,a1 and b1 from fruits were randomly distributed among genotypes.But lycopene content was negatively correlated with L2 and b2 of fruit puree respectively and positively correlated with a2.After free water in fruit puree was discarded by centrifugation,readings of L3,a3 and b3 from these pastes were more stable and displayed a more precise linear relationship to lycopene content.Free water imposed a significant effect on color readings.Among genotypes with free water content between 71.11% and 76.20%,the correlation coefficient between lycopene content and (a3/b3+a3/L3) was as high as 0.808.Therefore,linear regression equation could be used to predict lycopene content of fruits based on their color readings from paste without free water.

    The effect of SA on physiological indexes and gene expression were studied in tomato seedlings under low potassium stress in order to provide an important theoretical basis for the relationship between SA and low potassium stress. In this study,the different concentrations of salicylic acid (SA) (0.10,0.25,0.50,1.00 mmol/L) affect antioxidant enzyme activities and gene expression in tomato seedlings under low-potassium stress were investigated.The results showed that the chlorophyll (Chl) and proline (Pro) contents,as well as catalase (CAT),ascorbate peroxidase (APX),peroxidase (POD) and superoxide dismutase (SOD) activities,were significantly increased,and malondialdehyde (MDA) concentration decreased under 0.25 mmol/L SA treatment compared with that in the control.Meanwhile,the gene expression levels of APX,CAT,POD,and SOD in tomato leaves were analyzed through quantitative Real-time PCR.Four gene expression levels of antioxidant enzymes were considerably increased after 48,96 h treatment.These results suggested that SA promoted the gene expression levels of APX,CAT,POD,and SOD to alleviate the damage of low potassium stress,and decreased the accumulation of ROS and enhanced resistance in tomato seedlings under low-potassium stress.

    Using self-made LED light source the experiment study the effect of different light quality and light quality combination on morphogenesis and physiological and biochemical characteristics of tomato.The experiment accumulates data for application of LED light source in horticulture planting.The results showed that,the Blue and Green treats were all beneficial of improving the plant height of tomato seedling except the light quality treats were all beneficial of improving the stem diameter and fresh weight and dry weight and of tomato seedling except Red and Yellow treat and the healthy index of RB3 and RBP2 were the highest.They can be looked as the best light for growing tomato healthy seedling.All treats didn't improve the activity of CAT.The light quality treats RBG2 and Y and RB4 and RBP1 and RBG1 and RBP2 were all beneficial of improving the activity of POD.The light quality treats RBY1 and RB5 and RBY2 and RBP2 were all beneficial of improving the activity of SOD.Comprehensive evaluation,RBP2 can be looked as the best light for improving the activity of enzyme.

    Micro-Tom is a miniature dwarf tomato,which can grow at a high density, has a short life cycle, and can be transformed efficiently. As a result, it became a new model p lant for functional genomics study. Co-cultivation time on regeneration, effects of AS on the regeneration frequency, Agrobacterium titer and GFP exp ression, antibiotic concentration selection were studied to establish stable p rotocol. It indicated that the average T2DNA inserted sites were 118 and positive rateswere 7213% through PCR detection.

    This experiment consists of eight varieties of tomato were collected from home and abroad,the indexes of physiological and biochemical characteristics,field resistance,yield and quality were determined after the haze.The results showed that:during the weather of low temperature and haze,solar greenhouse's own environmental conditions far could not meet the requirements of the temperature,light and humidity of tomato.Under stress conditions,the varieties with stronger resistance were Jinpeng No.8 and Fenyan No.1,their fruit drop rates were 2.5%,3.7%,dead rates of plant were 38.8% and 25.0%,the diseases incidence rates were 35.5% and 35.0% respectively,These indicators were in the lower level than other varieties.And Jinpeng No.8 had the highest yield,54 300 kg/hm2,followed by Fenyan No.1and T1701,which production were more than 50 000 kg/hm2. Fruit quality,the maximum of sugar acid ratio was Jinpeng No.8,and the taste was best;The minimum of the nitrate content was Fenyan No.1.The whole fog and haze,the smallest decrease amplitude of chlorophyll was Jinpeng No.8,Fenyan No.1 wasthe second.Physiological and biochemical indexes,the varieties of accumulated more contents of soluble sugar and proline were Jinpeng No.8,Fenyan NO.1,the value of SOD and POD were also higher,The content of MDA didn't have obvious differences between species in this experiment.Therefore,under low temperature and weak light,the varieties with self-regulation,strong adaptability were Jinpeng No.8,Fenyan NO.1,which results of physiological indexes were consistent with the results of field experiments.

    The study focused on the influences of mushroom residue,peat and vermiculite matrix product on the growth characteristics of tomato seedlings.After matrix treatment,mushroom residue was mixed with turf and vermiculite for tomato seedling culture.The compound matrix mushroom residue treatment products were added with different proportions:0,33%,50% and 67% respectively,and the survival rate and growth characteristics of the tomato seedling were tested 25,45,65 days after sowing.Statistical analysis showed that the survival rate of tomato seedling with compound matrix T1,T2 and T5 reached 100%,3.3% higher than CK,the survival rate of tomato seedling with T4 and T5 were lower than CK 23.4% and 33.4% respectively.After comparison of 45 d tomato seedling height,stem diameter and root development indicators,those of T3 and T4 were significantly lower than those of CK,T1,T2 and T5.However,with the growth of the tomato seedling,at 65 d root lengths of T3 and T4 were significantly higher than those of CK,seedling height and stem diameter had no significant difference with those of CK,and the stem diameter and root lengths of T1,T2 and T5 showed no significant difference with CK,and the seeding height were higher than those of CK.The study indicated that the growth index of treatment T1(mushroom residue: peat: vermiculite=1 : 1 : 1) was higher than other treatments,and a relative high additive proportion of mushroom residue may inhibit the initial growth of tomato seedlings to some extent,which may further influence the accumulation of seedling biomass above the earth,yet as the seedlings keep growing,such inhibition was decreasing.At the later development stage of the seedling,increasing the additive proportion of mushroom residue can further promote the development of the tomato seedling root and enhance the biomass accumulation of the plant.

    Chilling-sensitive tomato Money Maker and chilling-tolerant wild tomato LA1777 were studied,and effects of low night temperature of 15,9 ,6 ℃ and recovery on the resistant physiological index of tomato fruits were investigated. The results showed that soluble sugar,proline content and POD activities increased in tomatoes LA1777 and Money Maker after low night temeperature treated,but the variation in LA1777 is greater. SOD activity in LA1777 was lower than control,CAT activities was higher than control,otherwise, the SOD activity in Money Maker was higher than control,CAT activities was lower than control. The increase of MDA content in Money Maker was significantly greater than control,while no significant change in LA1777. These results demonstrate that the two types of tomato fruits exerted higher level of soluble sugar and proline content to chilling stress under short-time low temperature, the chilling-tolerant tomato exerted higher level of POD and CAT activities to chilling stress,while the chilling-sensitive tomato exerted higher level of SOD activity, the enhanced membrane lipid peroxidation in the chilling-tolerant tomato fruits is lower.

    In order to prepare the special antiserum of Tomato chlorosis virus (ToCV),the total RNA of tomato leaves infected with ToCV were extracted.According to the coat protein gene of ToCV,specific primers were designed to amplify the coding region of coat protein by RT-PCR,the prokaryotic expression vector pET32a-ToCVCP was constructed and the recombinant protein was expressed in E.coli Rosetta (DE3).The results showed that the ToCVCP gene(NCBI:KT809400)owned 774 bp nucleotides,encoding 257 amino acids.The similarity of nucleotide and predicted protein were 97.2%-99.6% and 97.3%-100.0%,respectively,compared with the CP gene of other ToCV isolates registered in GenBank.The nucleotide sequence of conserved sites accounted for 91.3% of all loci,the amino acid sequence conserved sites accounted for 88.3% of all loci,indicated that the geographical origin of different ToCV CP gene had a relative higher conservative property.The ToCV CP gene was subcloned into the expression vector pET-32a(+) and the fusion protein was expressed in vitro.SDS-PAGE analysis showed that a specific recombinant protein of approximately 33 kDa was produced in the Rosetta (DE3) with the prokaryotic expression vector pET32a-ToCVCP in 37℃ with 1.0 mmol/L IPTG for 6 hours.

    Five invertases were investigated by semi2quantitative RT2PCR in different developing stages of the fruit, using genotype tomatoes,″ Momotaro″. The invertase genes expressions of tomato seedling under sub2high temperature,sub2 low temperature,NaCl salinity stress and osmotic stress were further studied. The results suggested that the expanding pe2 riod of fruit is affected by the expression of Lin5,Lin6 and Lin8.Only Lin8 expressed in mature green stage,while Lin6, Lin8 and AI2 expressed distinctly. The results showed that different members of acid invertase gene family regulated the different periods and tissues of tomato fruits development. Suboptimal temperature had significant influence on AI2. Whether sub2low temperature or sub2high temperature,both of them inhibited gene expression significantly,however,they have little effect on the expression of the cell wall2binding acid invertase genes (Lin5,Lin6 and Lin7) and have inhibited effect on Lin8. Lin5 and Lin7 have no expression in osmotic stress and salinity stress;however,the expressions of Lin6 and Lin8 are inhibited by osmotic stress and salinity stress. The expression was low when treated for three days,and for five days it was totally inhibited. AI2 is sensitive to osmotic stress whose expression is totally inhibited when treated for three days,meanwhile under prolonging the time of the salt stress,the expression is low,then completely disappeared.

    To clarify the suitable application rate of the soluble ammonium polyphosphate fertilizer for tomato,the phosphorous rates of soluble ammonium polyphosphate fertilizer on yield,quality,phosphorous uptake of autumn-winter tomato were studied in a plastic greenhouse.The results showed that the yield of tomato and the fruit weight were highest at the 29 kg/ha,however,when continue to increase the amount of phosphorus,the yield and fruit weight will be lower.The most appropriate phosphorus rate for tomato quality was 29 kg/ha.As the phosphorus uptake of tomato roots,the difference between treatments was not significant; After fitting,the most appropriate phosphorus rate for tomato fruit and whole-plant were 21.17 kg/ha and 25.79 kg/ha respectively.The phosphorous utilization efficiency (PUE) was higher than other treatments at 29 kg/ha.In summary,the most appropriate phosphorous rate for a higher autumn-winter tomato yield was 29 kg/ha under this soil fertility.

    In order to optimize Cd low accumulation or Cd high resistant tomato varieties,in this paper,4 tomato varieties(Provence,Ruitina,Kate 2 and 908) were selected as the research objects,and the differences of growth index and resistance index of tomato seedlings under cadmium stress were compared in detail by hydroponics experiment. The results showed that 908 had the highest natural characteristics in plant height,root length,total biomass,MDA and chlorophyll content under the condition of nutrient solution without Cd;Provence showed the most prominent in CAT activity;Ruitina showed the highest activity on POD. Under the condition of Cd stress,low Cd content promoted the growth of root system of Provence,which was 24.72% higher than that of CK,other varieties did not change much;the plant height of Ruitina was most sensitive to the stress of middle and high Cd content,other varieties had no obvious change. CAT of four tomato varieties under low and medium Cd stress has increased 12.58%-84.91%;the activity of CAT decreased sharply and the resistance function was seriously weakened under high Cd stress. The resistance of POD in 908 to medium and high Cd stress was increased,and the enzyme activity was enhanced by 76.07% and 72.65%;the activity of POD in Ruitina decreased sharply and the resistance was seriously affected. The contents of Cd in shoots and roots of 908 were the highest among the four varieties,3-12 times of other varieties,Provence was the lowest,and the transport coefficient of four varieties were all<1. Under the same Cd stress,the high Cd concentration absorbed by the 908 plants induced a high increase in the activity of antioxidant enzymes,reflected that the scavenging ability of 908 plants to reactive oxygen species was stronger than that of the other three varieties. In conclusion,under the medium and high Cd stress,Provence was selected as Cd low accumulation tomato variety,908 had the strongest resistance and higher Cd content,belonging to the high Cd resistant varieties.

    There exists the compound infection of Tomato chlorosis virus (ToCV) and Tomato yellow leaf curl virus (TYLCV), and the incidence of the two diseases presents a tread of increase in recent years, which becomes a new threat to tomato farming. In order to study the interaction and variation of the two virus genes after single and mixed infection, the molecular identification and sequence analysis of the CP gene and the HSP gene of ToCV and the gene of TYLCV occurred in Tianjin were carried out. The results showed that, compared with the single infection samples, 6 base mutations of TYLCV were found in mixed infection samples, i.e. 73rd A→C, 92nd A→G, 347th C→T, 1 209th C→T, 1 618th A→G and 2 107th A→T, among which the 73rd and 92nd mutations were not in the coding region and the other four were located in the ORF box with 3 missense mutations and one samesense mutation(1 209th). There was also one samesense mutation in the CP gene of ToCV. There were 4 base mutations in HSP gene of ToCV, among which the 826th T→C and 1 166th G→A were samesense mutations, while the 1 395th A→G and 1 630th A→G were missense mutations. The accumulation amount of TYLCV and ToCV in tomato samples after single infection and mixed infection were analyzed by RT-qPCR. The results showed that the accumulations of TYLCV and ToCV had no significant difference between single infection and mixed infection. In the mixed infection samples, the copy of TYLCV was about 262-436 times that of ToCV.

    Tomato scions(Zhongza 105,Zhongza 109,Yingfen 8)and rootstocks(Chongqing TR,Guizhen 1,GuiTA 06)as the materials,the physiological and morphological indexes of tomato hypocotyl were investigated after fertilization and uniconazole.Propose technology of fertilization and uniconazole composited application to optimize growth of tomato scions and rootstocks in grafting.The results showed that fertilizer treatment can improve tomato hypocotyl and epicotyl length and weight,and reduce the lignin content;Uniconazole spraying treatment inhibits epicotyl length and strench,and promote dry weight,the lignin content and anthocyanin of hypocotyl.Fertilization and uniconazole can effectively regulate the growth and development of tomato hypocotyls.

    Fruit carotenoid levels in tomato could be elevated significantly by high-pigment genes hp1 and hp2.They are valuable for quality breeding program targeting increasing lycopene content in tomato fruit.Based on their point mutations,specific molecular markers correspondent to them have successfully developed.Then,hp1 and hp2 could be accurately tagged by these markers,as the markers are in fact amplificated products of gene parts.In F2 population the segregation of molecular markers were in consistent with expected ratio of two independent genes.Lines with both hp1 and hp2 have been developed based on the molecular markers.

    In the summer of 2014,tomato plants showed suspected symptoms caused by Tomato chlorosis virus (ToCV) infection collected in greenhouse in Tianjin.In order to clarify the Pathogen of tomato plants,specific primer pairs of ToCV coat protein (CP) and heat shock protein 70 (HSP70) were used for reversing transcription PCR (RT-PCR) detection,specific fragments were obtained and sequenced.HSP70 nucleotide sequences obtained 99.5% similarity with ToCV isolate Tochigi (AB513442),CP amino acid and nucleotide sequences obtained shared 99.2% and 99.3% similarity with ToCV isolate Tochigi (AB513443),indicating the tomato was infected by ToCV in Tianjin,the sequences of ToCV isolate between Tianjin and Japan with a highest similarity.As Tomato yellow leaf curl virus (TYLCV) were widespread in survey areas,the ToCV positive samples were also tested for TYLCV by specific primers.The result was positive and gene sequence analysis showed that the similarity between amplification products and TYLCV-IL isolates was more than 99.0%,indicating mixed infections in the field.It is the first report that ToCV infect tomato,ToCV and TYLCV mixed infections tomato in Tianjin.

    The contents of different sugars(sucrose,fructose,total soluble sugar and starch) and the activities of sucrose phosphate synthase,sucrose synthase acid invertase,neutral invertase involved in sucrose metabolism was measured in order to know the sugar metabolism in developing tomato fruit.According to the development and the relative growth rate of tomato fruit,we can devide the tomato fruit development into two PHASes.The first PHASe before 39 days after anthysis forms the main output and in the PHASe after this the inner quality forms.In the first PHASe the contents of fructose and soluble total sugar increase gradually,the content of starch decreases and sucrose's content changes little.At the same time the activities of sucrose synthase and sucrose phosphate synthase relatively high,the invertases'activity relatively low.In the second PHASe,the content of fructose increase,the starch's content decrease and the contents of total soluble sugar and sucrose change little.The activities of invertases rise acutely,but the activities of sucrose synthase and sucrose phosphate synthase maintain very low and change little.By statistic analysis,we find the single fruit weight and the content of starch have significant correlation,while the content of starch has significant correlation with neutral invertase,so we can consult the content of starch and the activity of neutral as the index to gain high output.The contents of fructose、total soluble sugar and starch have the significant correlation,the contents of fructose and the activity of SS,AI,NI have significant correlation.The contents of total soluble sugar and the activity of sucrose synthase have significant correlation,so we can consult the activity of synthase and invertases as the index to gain high contents of fructose.

    In order to study the function and molecular mechanism of SlYAB2a in growth and development of tomato.The cDNA sequence of SlYAB2a was cloned from tomato by RT-PCR.Bioinformatics analysis showed that SlYAB2a encodes a putative polypeptide of 192 amino acids with a calculated molecular mass of 21.35 kDa,a theoretical pI of 8.79,and a grand average of hydropathy value of-0.487.SlYAB2a protein was predicted to possess a Pfam:YABBY domain architecture at position 6-165,a C2C2 zinc finger-like domain at position 20-47,and a helix-loop-helix domain at position 122-181.Secondary structure analysis revealed that SlYAB2a protein contains 19.79% α-helical domains,14.06% β-sheet domains,and 66.15% random coil.The analysis of protein multiple sequence alignments showed that the percent identity of the SlYAB2a with members of group YABBY2 from Solanum tuberosum,Nicotiana sylvestris,Vitis vinifera,Glycine max,Arabidopsis thaliana,Brassica napus,Oryza brachyantha,Zea mays and Triticum aestivum were 98%,78%,71%,65%,64%,64%,58%,57% and 57%,respectively;which suggested the percent identity of the SlYAB2a with other members of group YABBY2 from dicotyledon was higher than that from monocotyledon.The analysis of phylogenetic relationship showed that YABBY2 was easily separated into two distinct subgroups,that was dicotyledon subgroup and monocotyledon subgroup.SlYAB2a belonged to dicotyledon subgroup and was most closely related to members of group YABBY2 from Solanum tuberosum,Solanum pennellii,Nicotiana tomentosiformis and Nicotiana sylvestris. The expression of SlYAB2a was determined by Real-time quantitative RT-PCR.The result showed that the SlYAB2a gene was transcribed in different tissue organ.The expression level of SlYAB2a in flowers and mature green fruits was much higher than that in any other analyzed tissue organs.These results suggested that SlYAB2a might be involved in flower and fruit development.Following transient expression of SlYAB2a in onion epidermal cells,SlYAB2a was found to be localized in the nucleus.

    The effects of exogenous H2S and H2O2 physiological regulation on the processing tomato seedlings under NaCl stress were investigated,KT-32 with weak salt tolerance and KT-7 with strong salt tolerance as the materia. Research of 50 μmol/L H2S and H2O2 were determined by pot culture under the conditions of 0.5,1.5,3.0 mmol/L. Effects of cotreatment on the growth,osmoregulation,material accumulation and reactive oxygen metabolism of seedlings of processed tomato under 250 mmol/L NaCl stress.The results showed that the cotreatment of exogenous H2S and H2O2 alleviated the inhibitory effect of NaCl stress on the growth of seedlings of processed tomato seedlings,and maintained a strong photosynthetic pigment content,relatived water content and root activity in processed tomato seedlings.And decreased the electrolyte leakage rate,the content of MDA,the content of H2O2 and the production rate of O2· free radical and enhanced the activities of antioxidant enzymes SOD,POD and CAT activity. Among the two strains,soluble sugar increased 24.0% and 29.4% by compared with 50 μmol/L H2S treatment alone at 50 μmol/L H2S and 1.5 mmol/L H2O2 ,proline content increased by 25.8% and 21.3%,respectively,and the effect of salt stress alleviation was the most significant.In summary,there were a certain degree of synergy between exogenous H2S and H2O2,which could enhance the osmotic regulatory capacity of plants under enhanced NaCl stress in tomato seedling plants to enhance the removal of reactive oxygen species H2S and H2O2 in the tomato seedlings,and then weakened the injury of reactive oxygen species on the plasma membrane in processed tomato cells and then increased the adaptability of tomato seedlings to salt stress. The synergistic effect of the two signaling molecules will provide the theoretical basis for alleviating the physiological mechanism of salt stress in tomato seedlings.

    Heavy metal pollution in agricultural land becomes more and more serious in recent years. The soil culture experiments were performed to study the effects of combined pollution of Cu,Zn } Pb and Cd on growth of tomato and heavy metal accumulation regularity. The results showed that with the increasing of soil heavy metals Cu,Zn,Pb and Cd,the content of Cu,Zn,Pb and Cd in root of tomato were obviously increased. Zn and Cd content stems and leaves were increased. Cu and Pb content in stems and leaves were and Cd content in fruits were increased slightly. Cu,Zn,Pb and Cd uptake in not obviously increased,Cu,Zn, stems and leaves of tomato were far greater than that of fruits and roots. When the soil heavy metals content were higher Cu and Pb uptake in the stems and leaves decreased due to the decreasing of biomass. With the increasing of soil heavy metals content,Cu,Zn and Pb uptake in leaves of tomato were firstly increased and then decreased. With the increasing of heavy metals content in soil,Cd uptake in stems and leaves in tomato was rising trend. With the increasing of soil heavy metals content Cu,Zn,Pb and Cd uptake in fruits were not changed significantly,Cu and Pb uptake in root was always increased,while Zn and Cd uptake in root changed slightly. In the same soil heavy metal processing conditions,the accumulation ability of heavy metal elements in each part of tomato was Cd>Zn>Cu>Pb. The distribution of heavy metals of Cu,Zn,Pb and Cd in different parts of tomato was root>stem and leaf>fruit.

    Comparative observations on microspore development in a male sterile 95305 and fertile plants of L.esculentum were made with light and transmission electron microscopy. The results showed that the microspore abortion occurred at the stage of microspore mother cell, tetrads and uninucleate microspore. The tapetal cells were desolved completely earlier than that of normal male fertile material, or divided into multilayer to press the microspores. Other kinds of microspore abortion such as the failure of exin deposition also were reported. In addition, some anthers were deformed and had pistillody.

    The effect of light quality on the photochemical activity of tomato fruit surface was studied by a MINI-IMAGING-PAM system. The results showed that as the intensity of light quality rises(200-2 500 μmol/(m2·s)), the maximal photochemical efficiency(Fv/Fm), the effective PS II quantum yield(Y(II)), the apparent electron transport rate(ETR) and the open PSII reaction centers(Fv'/Fm') of tomato fruit surface which was under red light and blue light treatments all decreased in the shape of " S", while the relative deviation(β/α-1) from full balance between two photosystems rised gradually, indicating that excitation energy high intensity has broken the balance of excitation energy distribution between PSII and PSI, thus leading to the poorer cooperation of two photosystems. Chlorophyll fluorescence of tomato fruit surface which was under green light treatments was proved to be feasible, showing that green light may reduce the chlorophyll to absorb excessive of light energy and decrease photoinhibition. Fv/Fm、Y(Ⅱ) 、ETR and Fv'/Fm'of tomato fruit surface which was under blue light was smaller than red light and blue light, these gaps will widen with an increase in intensity, which indicated blue light was sensitive to photoinhibition. In case of lower light intensity(200-1 000 μmol/(m2·s)), Fv/Fm、Y(Ⅱ) and Fv'/Fm'of tomato fruit surface which was under red light was higher than green light, while over 1 000 μmol/(m2·s) leading to reverse results, which showed that the capacity of adapting to high light intensity of tomato fruit surface which was under green light was strong.

    In order to study the interaction between powdery mildew fungus and tomato, the AD-cDNA library of tomato leaves infected by Oidium neolycopersici was constructed using SMART technology.The titer of the constructed cDNA library was 3.77×1010 cfu, and the recombination rate of the library was about 96%, the insert fragments ranged from 400 bp to 2 000 bp, and the average length of the library was about 1 000 bp.Fifty clones were randomly picked from the library for sequencing and functional analysis.The results indicated that the library was qualified for the study on the interaction between powdery mildew fungus and tomato.

    Using single factor randomized block experiment,make saline water (EC=3 mS/cm)irrigation directly as control,to study effect on contents of mineral elements of tomato plants'roots,stems,leaves at fruit-set period,the early stage of full bearing period,later stage of full bearing period and different growing periods fruits of different irrigation methods of saline water.Results showed that:①These ten kinds of mineral elements in tomato roots,stems,leaves and fruits all showed regular changes.Six kinds of mineral elements contents in roots,stems,leaves were N,K,Ca greater than P,Mg,Na,in four kinds of microelements,the Fe,Mn was higher,next was Zn,Cu was the lowest;Except K,young fruits had the highest contents of five kinds of macroelement,the content of K was the highest in mature fruits.Microelements' content in fruits were Fe,Zn was higher,Cu,Mn was lower.In fruits,K was positive correlation with the other nine kinds of mineral elements.Na,Mg,Mn was antagonism with Zn,Cu,P. ② Saline water irrigation increased Na content of tomato root, stem, leaf and fruit in quality formation period, direct irrigation with saline water increased the maximum. Compared with freshwater irrigation, the total N content of all parts of tomato plant is also improved under saline water irrigation directly, but it was not conducive to the absorption and accumulation of N, P and 4 kinds trace elements of tomato fruit. Fresh water irrigation is beneficial to improve the content of P, K and Zn in each part of the plant, and promote the uptake and accumulation of N, P, K and Fe, Zn of tomato fruit. The content of Ca, Mg and Fe in each part of the plant can be improved when use saline water and fresh water rotation irrigation according to plant growth periods, Ca and Mg content of tomato at young fruit period and mature period are also higher under saline water freshwater irrigation in sequence and rotation irrigation according to plant growth periods. The contents of Cu and Mn in each part of plant are higher when use mixed water irrigation and saline water fresh water irrigation in sequence, so the content of Cu under mixed water irrigation and the content of Mn under saline water fresh water irrigation in sequence in the fruit are also higher at fruit quality formation period. So,using saline water irrigation in a short time or decreased the EC of saline water (rotation irrigation,mixing water irrigation)could avoid the negative influence of saline water and supplement the necessary elements of plants,they were N,Ca,Mg and Fe,Mn,Cu and so on.

    In this study,E6203(common tomato)and TA517(introgression lines of Solanum habrochaites)which have significant different lycopene content,were selected to be experimental materials. Specific primers had been designed, which based on GGPS fragment obtained from the subject group of fresh tomato in CAAS and the gene(DQ267903)publi2 cized in NCBI to amplify DNA and cDNA. Sequences analysis indicated that GGPS doesn′ t have intron. Putitive amino acid showed that 13 different residues are existed in the two GGPS,9 of them characters changed. The mutations lead to the difference of secondary structures,so do phosphorylation sites. It is suggested that the different lycopene content relat2 ed with amino acid mutations. 5′ 2flanking sequence were obtained by genome walking. Further analysis showed that it con2 tains significant transcriptional regulation sequences and the distribution of cis2acting element was conserved in two GGPS flanking sequence,but sequences had more difference. Semi2quantitative RT2PCR analysis presented that GGPS expressed mainly in fruit and flower.

    An advanced backcross population of 130 BC2S4 individuals was developed derived from the cross Solanum lycopersicum 9706 × Solanum cheesmanii LA0317.Six important phytological traits were investigated under salt stress and nonstress conditions,and salt survival value was also evaluated.Using MQM mapping method,a total of 45 QTL for phytological traits of tomato were detected.2 QTL for salt tolerance were mapped to chromosome 3 and 6.Through comparative QTL analysis,it was found that QTL on chromosome 2,3 and 8 could only be detected under salt treatment,while QTL on chromosome 9 and 12 could only be detected under non-salt condition.QTL co-location among different traits was common in this study.Moreover,5 families of BC2S4 were found in this paper to be salt tolerant,thus these lines could be used as good breeding germplasm.

    This paper studied effects under normal and low temperature of different phosphate fertilizer on tomato seedlings growth and physiology activity. The results showed that the phosphorus application (P2O5), ranging from 0.20 to 0.85 g/kg soil under ambient temperature could benefit to stronger tomato seedlings, when the phosphorus application exceed 0.85 g/kg soil the gowth of tomato seedlings begin to come down; under low temperature condition phosphorus application (P2O5) 3.14 g/kg, the growth of tomato seedlings reach the maximal value. Proper phosphorus application may improve tomato seedlings growth, the contents of chlorophyll and soluble sugar, but the contents of MDA were declined, that means the resistance ability improves; By the excess phosphorus application, tomato seedlings physiology activity reduces, the stress resistance was declined.

    The article tested and analysed the difference of the fruit firmness, respiratory intensity, lycopene, storage index and various commercial quality of the tomato ripening mutant material L2 and normal ripening cultivar L5 and their hybrid fruits L20 during storage process. The result shows that during storage process the change in fruit firmness of tomato submits downtrend, and the soluble solid, soluble sugar, Vc, lycopene content show uptrend, but the change in organic acid shows overall downtrend, so the ratio between sugar and acid shows uptrend; there is quite large difference among the respiratory peak, the fluctuation of L2 is relative small, and respiratory rate keeps the lowest all the time; the storage index of tomato fruit with rinis the highest, above 75%, but the storage index of normal fruit is the lowest only 60%, although the storage capability of their hybrid descends remarkably, the storage index is higher 15% than normal fruit.

    In order to identify the function of SlETR6 gene in tomato under different abiotic stresses, the SlETR6 gene was cloned by RT-PCR from Solanum lycopersicum, and the phylogenetic tree was clustered based on its amino acids by MEGA 5.0 soft. The temporal and spatial (included root, leaf, flower, and fruit in different stage) expression patterns of SlETR6 were analyzed by Real-time quantitative PCR (qPCR), and the relative expression level of SlETR6 gene was analyzed by qPCR under high salt stress, high temperature, low temperature and drought stress during tomato seedling development. The results showed that SlETR6 contained 2 265 bp ORFs, encoded 754 amino acid residues. The protein SlETR6 with a molecular mass of 80.05 ku and isoelectric point of 7.28, and had a closer relationship with Solanum tuberosum ETR2-like. Promoter analysis revealed that SlETR6 contained several stress-responsive elements involved in heat, drought, anaerobic, light, ethylene, salicylic acid, and gibberellin signaling. qPCR analysis showed that the SlETR6 was constitutively expressed in all examined tissues, including seed, root, flower, leaf and fruit,and it showed a higher level in flower and breaker fruit. SlETR6 was up-regulated under high salt stress 6 h compare to control, and the expression level was the highest at 12 h under stress, then return to the normal. The transcription level of SlETR6 was up-regulated significantly under high-temperature stress. Drought stress could significantly up-regulated the expression of SlETR6 in the early stage, and the expression level was the highest at 1 h under stress. These results suggested that SlETR6 might function as an stress-responsive gene under high salt stress, high temperature and drought stress during tomato development. This study provided a potential novel candidate for abiotic research in tomato.

    To obtain the convection between tomato fruitage and air, the differences in temperature of tomato fruitage surface and air on controlled environment temperature, light intensity in different wind speeds were measured. The convection coefficient between tomato fruitage and air was calculated according to heat balance law,and kc,ks parameters of the models were calculated using Porter and Mitchell's models. The influence of hidden heat coming from tomato water loss was analyzed through 24 h time interval weight loss of the tomato fruitage. The result showed the hidden heat of the tomato fruitage was 2. 3 W/m2. The measurement result of the convection coefficient increased according to the increase of wind speed,which confirms the view point of former researchers about convection coefficient. It gives a definite parameter for convection heat analyses. Not only did this paper provide a measurement method of the convection coefficient, it settled a base for tomato fruitage climate space study also.

    The content of Chla content,Chlb content,Chla/b value of Number4zhongshu and Number9zhongza in low light and poor light were studied. The research results showed that when light intensity is 5 000 lx or 2 000 Ix,temperature treatment 20℃(day)/10℃(night),20℃(day) /5℃(night ),15℃( day ) /10℃(night)and 15℃(day)/5℃(night) have a significant effect on Chla content,Chlb conten and Chla/b value of tomato, Which have different effect on different treat time.

    WRKY transcription factor is a family of transcription factors unique to plants.Studies have demonstrated that WRKY transcription factor played an important role in plant growth and development and in plant response to biological and abiotic stress.In order to reveal the function of tomato WRKY gene,two inbred lines of tomato with high resistance to bacterial wilt Hm 2-2(R)and high susceptibility to bacterial wilt BY 1-2(S)were used as experimental materials based on the preliminary transcriptome data,and a WRKY transcription factor SlWRKY75 gene(Solyc05g015850.3)was cloned.The structure,expression pattern and function of the gene and its encoded protein were analyzed by bioinformatics analysis,multiple alignment of amino acid sequences,phylogenetic tree construction,Real-time Quantitative PCR(qRT-PCR)and virus induced gene silencing(VIGS).The results showed that the full length of the cDNA of this gene was 653 bp,its maximum open reading frame was 519 bp,encoding 172 amino acids,the relative molecular weight of the protein was 19.878 51 ku,the theoretical isoelectric point was 9.32.The protein belonged to the hydrophilic non-secreted protein,and there was no transmembrane structure.Meanwhile,the protein had a highly conserved WRKY domain and a CX4CX23HXH zinc finger motif,which belonged to the Class Ⅱ family.Phylogenetic tree analysis showed that SlWRKY75 was closely related to Solanum pennellii SpWRKY75 and clustered into a group with other Solanaceae,while it was far related to Hevea brasiliensis HbWRKY75 and Gossypium hirsutum GhWRKY75 and was in different branches in the phylogenetic tree.The results of qRT-PCR analysis showed that the expression of SlWRKY75 gene were tissue-specific and could be induced by Ralstonia solanacearum,salicylic acid and jasmonic acid.VIGS result showed that silencing SlWRKY75 reduced plant resistance to bacterial wilt,indicating that SlWRKY75 positively regulated tomato resistance to bacterial wilt.These results suggested that SlWRKY75 gene played an important role in regulating tomato resistance to bacterial wilt.

    In order to compare and analyse the reasons of salt stress and water stress influence tomato yield and?fruit quality from the perspective of carbohydrate production and metabolism,the tomato variety Liaoyuanduoli was?used,the same osmotic potential of NaCl and PEG6000 was used to simulate salt stress and water stress,photosynthetic characteristics and sugar metabolism in tomato leaves of two treatment was studied. The results showed that:?Iso-osmotic NaCl and PEG stresses cause reduced net photosynthetic rate of tomato leaf,within 14 d PEGl treatment?reduced more than NaCl treatment,after 14 d NaCl treatment reduced more than PEG treatment,but,overall,little?difference between the two treatment;Iso-osmotic NaCl and PEG stresses reduced stomata conductance,intercellular?CO2concentration and transpiration rate in tomato leaves,increased stomata limitation value;Iso-osmotic NaCl and?PEG stresses reduced chlorophyll a,b and total chlorophyll content,decreased chlorophyll a/b in tomato leaves,little difference between two treatments. Under iso-osmotic NaCl and PEG stresses,the content of fructose and glucose?increased significantly in tomato leaves compared with control,PEG stress increased more than salt stress,sucrose?and starch content reduced in tomato leaves compared with control,water stress reduced more than salt stress;Isoosmotic NaCl and PEG stresses increased activity of sucrose invertase in tomato leaves,PEG stress increased more?than NaCl stress. The results show that:NaCl and PEG stress both destroyed tomato leaf photosynthetic function,decreased the efficiency of photosynthesis,but little difference between two treatments;NaCl and PEG stresses altered?the direction of sucrose metabolism in tomato leaves,increased significant the decomposition of starch and sugar,increased the content of fructose and glucose,the impact of PEG stress on sucrose metabolism was significantly greater?than NaCl stress in tomato leaves.

    To investigate the effects of organic and inorganic fertilizers on the yield and quality of pepper and tomato.Under the greenhouse cultivation condition,the pepper and tomato of field experiments were conducted to study the effect of yield and quality when reduced organic and inorganic fertilizers and combined the both application.The treatments were set up as no fertilizer,100% organic fertilizer,75% organic fertilizer,50% organic fertilizer,50% chemical fertilizer+50% organic manure fertilizer,75% chemical fertilizer,50% chemical fertilizer.The results showed that reduction and combined application of chemical and organic fertilizer had no significant effect on the yield of pepper, the effect of soluble solids content, soluble sugar content and soluble protein content was relatively small.The treatment that combined application of chemical and organic fertilizer and the quality of pepper which the content of nitrate was the lowest and the content of vitamin C was highest.While reduced the fertilizer application would lead to a different degree of yield increase for tomato, and compared with other treatments,treatment of 50% chemical fertilizer+50% organic manure fertilizer reached the highest yield and the best quality which the nitrate content of tomato was the lowest, and the content of vitamin C and amino acid were the highest, and the difference of soluble solids content and sugar acid ratio was relatively small.

    Using polyethylene glycol PEG6000 with osmotic potential from-0. 2 MPa to-0. 4 MPa as simulating water stress,experiment on germination of seeds of eight tomato varieties was done,effections of osmotica stress on tomato seed germination were researched. The result indicated that the higher the osmotic concentration was,the greater degree of coercion stress on seed germination was,all the materials could not germinate under-0. 4 MPa osmotic potential,the relative germinative energy,relative germination rate,relative germination index,relative vigor index,embryo roots growth were inhibited in diferent degrees under-0. 2 MPa and-0. 3 MPa osmotic potential,germination drought resistance index (GDRI) under two kinds of concentration were positively correlated (r =0. 759* ). By cluster and correlation analysis of GDRI and the other indices,we thought that PEG-6000 with osmotic potential from-0. 2 MPa to-0. 3 MPa could be used for drought resistance identification,it cleared identification indexes system of drought resistance at germination stage,laid the foundation of comprehensively evaluated drought resistance of tomoto varieties.

    To clarify the suitable application rate of the soluble ammonium polyphosphate fertilizer for tomato,the nitrogen(N) rates of soluble ammonium polyphosphate fertilizer on yield,quality,N uptake and distribution,N balance and N utilization efficiency of autumn-winter tomato were studied in a plastic greenhouse.The results showed that the yield of tomato was the highest at 137.8 kg/ha and the fruit weight was the highest at 116 kg/ha.The most appropriate N rate for tomato quality was 116 kg/ha.As the N uptake of tomato roots and stems,the difference between treatments was not significant;the most appropriate nitrogen rate for tomato fruit and whole-plant were 71.39,84.39 kg/ha respectively.The nitrogen utilization efficiency(NUE) was higher than other treatments at 116 kg/ha.For this treatment, the apparent loss of nitrogen was 113.75 kg/ha.In summary,the most appropriate nitrogen rate for a higher autumn-winter tomato yield was at 116 kg/ha and the NUE was 27.6% under this soil fertility.

    The purpose of this study was to determine whether the tomato plants in Tianjin was infected by Tomato chlorotic virus (ToCV) and whether variation had occurred in the important genes of ToCV.ToCV isolated in Tianjin was diagnosis at molecular level.The genome sequence of coat protein and heat shock protein had been cloned and analyzed.The results showed that the homology of the CP isolate in Tianjin was 97.3% or more with the representative isolates of different countries and regions.The homology of the Hsp70h isolate in Tianjin was 97.2% or more with the representative isolates of different countries and regions.It is proved that Hsp70h and CP protein are the most conserved proteins.This is the first report of Tomato chlorosis virus occurrence in Tianjin.

    To figure out the physiological regulation mechanism of tomato cold resistance,we used two type tomatoes which one is cold resistance wild tomato LA1777 and the other is low temperature sensitive cultivated tomatoes LA2706 as our test materials to study the influence of low temperature on antioxidant metabolism and carbon assimilation. Results showed that under the stress of low temperature,the contents of superoxide-anion radical (O2·),hydrogen peroxide(H2O2),malondialdehyde(MDA),dehydroascorbic acid(DHA)increased,and it that in LA2706 was about 0.3%-21.0%,1.2%-17.3%,13.4%-36.9%,and 13.3%-21.0% higher than LA1777,that speeded up the process of membrane lipid peroxidation. The contents of reduced ascorbic acid(AsA)had a slight upward trend with cryogenic processing time extended;the activity of superoxide dismutase(SOD)decreased by 55.3% and 28.2% 7 d after treatment in LA2706 and LA1777,LA2706 fell obviously than LA1777;the peak of ascorbic acid catalase(APX)in LA1777 was significantly higher than LA2706,and about is 1.7 and 4.0 times that of LA2706 in 1 and 7 d after treatment. With the extension of low temperature processing time,the total sugar content,acid invertase activity,neutral invertase activity and sucrose phosphate synthase all raised,LA2706 had a significantly higher increment than LA1777,and about was 5.8,4.8,10.2 and 1.7 times in 7 d that of 0 d after treatment.Low temperature made LA2706 to have a large number of decomposition starch into sugar,to alleviate the stress of low temperature environmental damage.

    In this paper, taking peat, perlite and vermiculite as the basic experimental material, established dif-ferent physicochemical properties of compound substrate through the way of different proportion of basic materials.Applied regression analysis and path analysis to study the indicator of substrate physicochemical properties on toma -to plug-seedlings,in order to achieve the standardization of substrate physicochemical properties and screen sub -strate formula of tomato seedlings production.The result indicated that, peat∶perlite∶vermiculite=6∶4∶1 ( V/V) was best for tomato plug seedlings ( Physicochemical properties as follows:Bulk density was 0.113 g/cm3, total porosity was 64.65%,air-filled porosity was 40.22%,container capacity was 24.44%,pH-value was 6.19,EC-value was 0.76 mS/cm,available nitrogen content was 386.9 mg/kg,available phosphorus content was 211.5 mg/kg,availa-ble potassium content was 826.5 mg/kg).Through the comparison of seedling morphological index and quality in-dex, we found that tomato seedling cultivated with peat∶perlite∶vermiculite=6∶4∶1 was significant higher than oth-er substrate formula on stem diameter and strong plug-seedlings index ( G-Value,root-shoot ratio, sound seedlings in-dex ).Regression and path analysis showed that direct path coefficient, positive and negative effects were different a-mong substrate physicochemical properties and tomato strong plug-seedlings index.Bulk density was the main physi-cal properties that influenced tomato strong plug-seedlings index in the production of tomato seedling.When the sub-strate physical property was in the optimum range.pH-value,available nitrogen,available phosphorus and available potassium content were the main factors which effect tomato strong plug-seedlings index.In order to got the best composite substrate formulation and cultivated strong and high-yield tomato seedlings, in the preparation of sub-strate, bulk density was the first consideration, within a suitable range, then adjusted pH value and available nutrient content for the requirements of tomato plug-seedlings.
