Relationship between the Neutralization Index in Chicken Embryo and the Homologies of HA Gene of Different H9N2 AIV Isolates

Abstract: In this paper,22 prevailed H9 N2 AIV isolates in China from 1998 to 2011 were identified,purified, sequenced,analyzed,and compared with the 46 H9N2 references in GenBank.Then,the phylogenetic tree was re-constructed based on those H9N2 strains.First of all,6 representative isolates were chosen,which belong to different genotype,to make for signal positive serum. Secondly,the cross neutralization test was determined by using positive serums in specific pathogen free(SPF)chicken embryos,respectively.Thirdly,the correlation of the virus neutraliza-tion index(VN)in chicken embryo was compared with the amino acid homologies of HA gene of different H 9N2 iso-lates by SPSS 14. 0 software in order to explore the relationship between antigenic variations and genetic evaluations of the prevailed H9 N2 isolates undergo the effect of vaccination. Importantly,the correlation coefficient and index of VN and amino acid homology was significantly correlated (P<0.01,r=0.639 8),which suggested that the H9N2 virus undergo more than ten years of vaccination,has changed to some degree at the molecular level and antigenici-ty.Collectively,these data implied that antigenic variations of H 9N2 isolates result in the genetic mutations of HA, which was essential for developing the new vaccine.
