Cloning and Sequence Analysis of a cDNA Encoding ACC Oxidase from Fuyu Persimmon

Abstract: Total RNA was extracted from Fuyu persimmon fruits and reverse transcribed to first strand of cDNA. A pair of primers were designed and synthesized based on the sequence reported. The specific PCR product of 1-aminocyclopropane-1-carboxylic acid oxidase was obtained using the first strand of cDNA as template. The product was ligased to pGEM-T easy vector and sequenced. The sequencing data showed that the PCR product was 1237 bp with 697 bp of coding region, encoding 231 predicted amino acid residues. Comparison with the cDNA sequence from DK-ACO1 indicated the homology was 99%, and amino acid identities were 98%.
