Temperature Influence on Transformation Efficiency Agrobacterium tumefaciens

Abstract: In this article,five co-cultivation temperatures (19,22,25,28 and 31 ℃) were evaluated to determine their effects on rape and maize Agrobacterium tumefaciens transformation efficiency. Rape hypocotyl and maize immature embryo explants were co-cultivated with Agrobacterium tumefaciens LBA4404 containing plas-mids encoding resistance to the herbicide phosphinothricin and chitinase gene. Explants were selected anti-cullus on medium with herbicide phosphinothricin. The results showed that, temperature has a large effects on transformation of rape and maize, Co-culture at 25 ℃ led to the highest number of stable transformed cullus. Although, there have remarkable diversity between 25 ℃ and 19 ℃, 22℃, 28℃ and 31 ℃ on-maize transformation efficiency, on rape transformation efficiency 25℃ and 22℃ have not remarkable diversity.
