Study on Rapid Propagation of Grape in vitro

Abstract: Experiments were conducted to study various factors affecting rapid propagation of grape (America red grape and America black grape) in vitro. Selection and sterilization of plant material, induction of shoot bud and multiple shoots, shoot rooting and plant transplanting were all studied. The original shoots from grape in June, July and August showed good potential for culture. The survival rate of axillary buds was higher than that of the apical buds. By dipping the original shoots in 75% ethanol for 20 seconds, and then treating with 0.1% mercuric chloride for4 minutes, the survival rate of shoot buds was up to 70%-80%. The 0.5mg/L6-BA solution significantly stimulated budding and shoots elongation, the propagation coefficient was3-4 times per week. Shoot rooting rate reached 90% when shoots were planted in the root-induction medium NAA (0.2mg/L). The survival rate of transplanted plants was around 80%, when they were conducted step by step.
