Studies on the Callus Type and Cytology ofthe Growing Branch of the “Xiao Zao” Jujube

Abstract: It was found in the in vitro culture of the callus from the growing branch of the “Jinsi Xiaozao” jujule ,that stimulants at different concentration and lighting significantly affected the quality of the callus.The use of 2,4 D in dark culture helped the induction of loose type Ⅱcalli,while IBA and lighting helped the formation of compact type I calli .The type I calli showed no capacity of differentiation ,while the type Ⅱcalli was found stronger in differentiation .During propagation 2,4 D at a concentration higher than 1mg/L resulted in the massive conversion of type I into type Ⅱcalli .Calli were found different in the variability of chromosome ploidy .The cells from the root tip of the regenerated plantlets were found relatively stable in their chromosome ploidy .
