Bioinformatics and Expression Pattern Analysis of DfTCP in Dryopteris fragrans

Abstract: In order to explore the response mechanism of DfTCP gene of Dryopteris fragrans to exogenous hormones and abiotic stress, the seedlings of Dryopteris fragrans were treated with exogenous hormones (SA,ABA,MeJA and Eth),salt (200 mmol/L NaCl),drought (20% PEG),high temperature (42℃) and low temperature(4℃),samples were taken at 0,0.5,1,3,6,12,24,48 h respectively,bioinformatics analysis and expression pattern analysis were carried out for DfTCP. The results showed that the total length of CDS of DfTCP gene was 1 431 bp,which encoded 477 amino acids. The expressed product contained a conserved bHLH domain,which was located outside the cell;the secondary structure of DfTCP was mixed type;the relative expression of DfTCP in different tissues of Dryopteris fragrans was petiole > leaf > root;under the treatment of exogenous hormone SA,the relative expressions of DfTCP in the leaves of Dryopteris fragransshowed a trend of "ascending-descending-ascending-descending",they reached the peak value at 0.5,12 h,which were extremely significantly higher than the control;under ABA treatment,the relative expressions of DfTCP in the leaves of Dryopteris fragrans were up-regulated and showed a trend of increased first and then decreased,but there was no significant difference between 0.5 h and the control group;after MeJA and Eth treatment,the relative expressions of DfTCP in the leaves of Dryopteris fragransshowed a trend of "descending-ascending-descending" with the increase of treatment time,reached the peak value at 6,1 h respectively,and the difference was extremely significant compared with the control;under the condition of PEG and NaCl treatment,the relative expressions of DfTCP in the leaves of Dryopteris fragransshowed a law of "drop-rise-drop" with the extension of treatment time,and reached the peak value at 6,12 h respectively,which were extremely significantly higher than that of the control;the relative expressions of DfTCP in the leaves of Dryopteris fragrans increased first and then decreased at different temperatures,and reached the peak at 0.5,3 h respectively at high and low temperature,which were extremely significantly higher than that of the control. In summary,the response patterns of DfTCP in the leaves of Dryopteris fragrans to different exogenous hormones were different,and DfTCP responded to different abiotic stresses at different time points,indicating that DfTCP played a positive role in the stress resistance of Dryopteris fragrans.
