CRYOPRESERVATION OF WINTER MULBERRY BUD

Abstract: The winter buds of mulberry were precooled at-3-20℃ and then by the rapid-frreezing method put into liquid nitrogen(LN,-196℃)without the use of a cryoprotectant.The winter buds were taken out from LN arid were thawed.To check the survival rate of winter bud the tissue culture method was used.The result was that survival rate was 55-90%.A better thawing method is 0-2℃ air slow-thawing.After storage for 1 to 40 days in LN,there is no difference in winter bud’s,survival rate.Buds could be normally differentiated arid developed through tissue culture.The results offer possibility for long duration preservation of winter mulberry bud.
