Isolation and Identification of Endophytic Bacteria in Tomato and Its Tolerance on High Temperature and High Salinity

Abstract: In order to screen tomato endophytic bacteria strains with good biological control effect and application value. In this study,an endophytic bacterium named JZ3 with high antagonistic activity against tomato grey mold was isolated. Through the comparison of morphological characteristics,phylogenetic analysis of 16S rDNA sequence and gyrB gene sequence,the results showed that JZ3 was a strain of Bacillus amyloliquefaciens. The results of laboratory test showed that,the preventive effect of JZ3 liquid suspension(5×107cfu/mL) on the tomato gray mold was up to 82.04%,the treatment effect was about 30.54%. The high temperature and high salinity tolerance of JZ3 was detected,the results showed that the survival rate of JZ3 in high salinity environment was 70%,the survival rate of JZ3 under high temperature and high salt condition(15 min) was still up to 50%. The storage stability of JZ3 was tested,the results showed that number of functional microorganism remained unchanged after storaged in the room temperature for 12 months. In conclusion,the Bacillus amyloliquefaciens strain JZ3 isolated from tomato plants has good biocontrol and storage properties,it provides a good strain resource for the development of efficient microbial preparation,and has broad application prospects.
