Effects of Culture Substrates and Temperature on Artificial Germination of Puccinia triticina

Abstract: The objective is to obtain the optimum culture substrates and temperature for artificial germination of Puccinia triticina and afford good experimental materials for RNA-seq of P.triticina.We cultured the fresh FHJT urediniospores in dark and high humidity to perform the effects of six different culture substrates on germination of it.The result showed that there was significant difference in germination ratio and length of urediniospore germ tubes on the six culture substrates under optimum culture temperature 20 ℃.The longest tube that germinated on nonwovens was up to 465.85 μm.However,germ tubes on microfiltration membrane grew faster than that on others,while there was no significant difference in germination ratio on these six culture substrates after germination started 12 hours.Microfiltration is the optimum culture substrates for germination of urediniospores of P.triticina.
