ISSR Analysis of Genetic Diversity in Asparagus

Abstract: The genetic diversity of 43 asparagus cultivars collected from 5 countries was analyzed by ISSR method in order to provide more data reference for asparagus breeding.The results show that a total of 159 bands were amplified by PCR with 18 primers,among which 121 bands were polymorphic.The percentage of polymorphism was 76.10%.For these asparagus,the observed number of alleles was 2,the effective number of alleles was 1.444 3,the Nei's gene diversity was 0.265 4,and the Shannon's information index was 0.411 4.Calculated using the formula,the average polymorphism index content(PIC) was 0.669,ranged from 0.317 to 0.840.The genomic average genetic richness was 5.17.This indicates that the genetic diversity in the tested materials with high performance.The genetic similarity coefficient was analyzed,and a tree diagram was constructed by NTSYS 2.10e software.UPGMA cluster analysis showed that these cultivars could be categorized eight groups according to coefficient 0.77.The cultivars JK104 from Jiangxi,Pacific Peak from Spain and 082 from Shandong were respectively identified as different single group.In other group,the genetic similarity between the cultivars from American and Jiangxi,American and Holland,Holland and Beijing were relatively high.
