Cloning and Expression Analysis of Calcineurin Gene from Curvularia lunata

Abstract: Millet curvalaria leaf spot is a very important leaf disease of foxtail millet. To understand the function of the calcineurin signal transduction in C.lunata,the calcineurin gene was cloned and preliminary studied. The ho-mologous fragment of catalytic subunit ( ClCna) and regulatory subunit gene ( ClCnb) of C.lunata was obtained by degenerate PCR and the full length cDNA sequence of calcineurin gene in C.lunata was cloned by RACE methods. ClCna and ClCnb gene were highly homologous to calcineurin in other filamentous fungi. The deduced amino of ClC-na gene showed it had three conserved regions,which included calcineurin B binding region,calmodulin binding re-gion and auto-inhibitory domain. The deduced amino of ClCnb gene contained four calcium-binding regions. Conidia germination was significantly reduced by cyclosporin A (CsA)treatment in C.lunata.By qRT-PCR analyzing,the ex-pression of ClCna and ClCnb gene were induced by NaCl and sorbitol in C.lunata.The study showed ClCna and ClCnb gene play a role in the adaptation to change in a biological stress and regulation of conidial germination.
