Study on Transformation of Starch Branching Enzyme SBEIIb Gene into Maize Inbred Line

Abstract: The SBEIIb gene was transferred into maize inbred line Danhuang 25 by Agrobacterium tumefaciens mediated transformation.Factors influenced transformation efficiency was studied.The results showed that 9 days with subculture of callus,AS concentration in the bacilli concentration at 10 mg/L,20 minutes infection time and co-culture 60 hours were the optimal conditions.PCR analysis on four resistance plants demonstrated that three of them were positive and the foreign gene was integrated into maize genome.
