ISSR Analysis on 6 Color Potato Varieties

Abstract: To know the degree of genetic difference among 6 color potato varieties which are Heimeiren,Russet-01,Desiree,Qingshu168,Shuixihong and Sepihong,and provide evidence for potato germplasm evaluation and cross breeding,the polymorphism among the tested plants were analyzed based on ISSR molecular marker. The results showed that 151 bands of ISSR markers were amplified with 12 suitable primers,and the length of DNA fragment were ranged from 250 to 2 000 bp,and 136 bands were polymorphic,and the percentage of polymorphism bands reached 88. 2% . The genetic distance(GD) of 6 tested plants were ranged from 0. 438 8 to 0. 644 4,and the average of genetic distance was 0. 539 4. Using the GD value of 0. 55 as threshold,6 tested plants could be clustered into 3 groups: the first group including Desiree,Qingshu168 and Sepihong,the second groups including Russet-01 and Heimeiren,and the third groups only including Shuixihong,which genetic relationship among 6 color potato varieties are different.
