The Influence of Vary Number of RAPD Polymorphic Loci to Information Evaluation of Genetic Variation in Adzuki Bean

Abstract: RAPD (Random amplified polymorphic DNA) variation was assessed in 163 accessions of adzuki bean from China,Japan,Korea,Bhutan,Nepal and Vietnam.A "population" was constructed by 261 polymorphic loci detected by 27 RAPD primers.The method of entirely random sampling was adopted to analyze the regression relationship between the sizes of sample (allelic number) and genetic similarity index (Jaccard index).Regression equation for different number of primers,loci and Jaccard index was calculated out,and then the reasonable number of primers and loci under given precision can be decided by analyzing the tangent of the curve equation.The results were as follows:There exists significant difference among the S,CV and average of Jaccard index calculated from different number of primers. Regression equation for different number of primers,loci and the CV of Jaccard index was Y=a+b/X,and the determine index was above 0.95.Under the precision of 5%-10%,the reasonable sample should be 15-32 RAPD primers or 120-230 polymorphic loci.
