Study on Transformation Efficiency by Agrobacterium-mediated in Tomato

Abstract: Tomato(Lycopersicon esculentum Mill ) cultivars,Texuan-28,Taiyuan 823were used as the materials,and cotyledon and hypocotyls were cultured to optimize the transformation system via Agrobacterium.The results showed that the highest transformation efficiency was obtained when the explants were preconditioned on preconditioning medium for 2 d prior to infect with Agrobacterium (OD600= 013) for 6 min growing on the culture mediumof MS+ 210mg/L 6-BA+015 mg/L IAA.It was proved that the target NPTII gene had been integrated into the genome of regenerated plants by PCR analysis.
