Abstract: The fragment ofSRYgene containing entire encoding region from Liaoning cashmere goat genomic DNA was amplified by the polymerase chain reaction(PCR)with a pair of designed primers based on the sequence of goatSRYgene sequence from GenBank.The amplified fragment was cloned to the pMD18-T vector,and transformed to the bacteria DH5α.The identified positive clone with Liaoning cashmere goatSRYgene fragment was sequenced.The sequences iden-tities ofSRYgene encoding region of some artiodactyla from GenBank were compared with the Liaoning cashmere goatSRYgene fragment,and the phylogenetic tree was constructed by NJ method.The results are as follows.The entire coding region of Liaoning cashmere goatSRYgene is 723 bp,encoding a peptide with 240 amino acid residues,and it contains the HMG-box with 231 bp between position 190 and 420,encoding 77 amino acid residues(H⁶⁴-A¹⁴⁰).The corresponding nucleotide sequences in GenBank of goat,sheep,cattle,yak,water buffalo,sika deer,musk deer,and pig had identities of 100%,97.09%,89.21%,89.21%,88.38%,87.55%,86.16% and 68.04%,respectively,with the Liaoning cash-mere goat.The clustering based on the sequences ofSRYgene encoding region among species is generally in agreement with the classic taxonomic relationship.
