Primary Analysis on the Antifungal Activity of the Protein Produced by Bacillus subtilis NCD-2 against Verticillium dahliae

Abstract: Bacillus subtilis NCD22 was isolated from cotton rhizosphere and showed significantly control effect on cot2ton verticillium wilt. Characteristics of the crude antifungal protein produced by strain NCD22 was evaluated under differ2ent culture conditions. The result showed that the strain could produce more antifungal substance under the optimum con2dition(cultured in NB broth for 2d under 30 ℃ and pH 710). Precipitatesof the fermentation of strain NCD22 was collectedby salt out with ammonium sulfate and showed strong inhibition effect against Verticillium dahliae. The culture of the am2monium sulfate precipitation from the crude protein extracts by RNase treatment,the antifungal activity compared withcontrol was no significant difference,when treated with Proteinase K,Trypsin,the antifungal activity compared with thecontrol was significantly. The activity of the antifungal substrates were investigated under different temperatures. The anti2fungal activity of the substrate at 60 ℃was no significant difference when it compared with control,while the antifungalactivity decreased at 80,100 and 120 ℃and showed significant difference compared with the control. However,the crudeprotein extracts still showed the antifungal activity after heat treatmentsfor 30 min under 120 ℃. Results indicated that theantifungal extracts of Bacillus subtilis NC-2 were several substrates with different traits.
