Screening and Identification of Phosphate Solubilizing Bacteria in Hordeum vulgare Rhizosphere and Its Growth Promoting Effect

Abstract: In order to obtain efficient dissolving phosphorus and growth-promoting Bacteria of Hordeum vulgare rhizosphere,we took study on screening,P-dissolving characteristics,growth promoting effect and identified by sequence analysis of 16S rDNA of strain PSM,which provided the basis for the development of solubilizing microbial fertilizer. The sequence analysis of 16S rDNA results showed that PSM was Bacillus subtilis,which had strong ability of degradation of inorganic phosphorus effect. After 6 days fermentation at a temperature of 35 degrees in the medium with Ca3(PO4)2 as the sole phosphorus source,soluble phosphorus content reached to 449.1 mg/L. PSM had obvious effect on promoting growth of Brassica campestris L.,the adding of 5.0×108 cfu/g PSM had the best effect on increasing the biological yield and number of leaves significantly. Compared with the single application of compound fertilizer(CK1),fresh weight and dry weight was increased by 16.72% and 18.68% respectively. Compared with the single application of peat carrier(CK2),fresh weight and dry weight was increased by 11.77% and 11.70% respectively. PSM showed evident promoting effect on plant.
