Research on Differential Expressed Genes in Maize under Low-zinc and Zinc-sufficiency Conditions

Abstract: Suppression subtractive hybridization (SSH) was performed to isolate the differentially expressed cDNA fragments under low-zinc and zinc-sufficiency conditions. In the forward subtracted library, the cDNA of maize seedlings treated with low-zinc was used as tester and the cDNA of maize seedlings treated with zinc-sufficiency was used as driver. In the reverse subtracted library, the cDNA of maize seedlings treated with zinc-sufficiency was used as tester and the cDNA of maize seedlings treated with low-zinc was used as driver. The cDNA fragments differentially expressed under low-zinc and zinc-sufficiency conditions were collected in the forward and reverse subtracted library, respectively. Thirty clones were randomly picked to sequence in each library. The sequencing results shows: the function of isolated genes in the forward subtracted library mostly relates to disease resistance/defense, signal transduction, cell growth/division and so on. While the function of isolated genes in the reverse subtracted library mostly relates to cell/tissue generation, metabolism and so on.
