Subcellular Localization of ZmSPK1, A Protein Kinase from Maize, in Transgenic Arabidopsis

Abstract: To determine the subcellular localization of ZmSPK1,a protein kinase from maize,using 35S promoter we constructed two plant expression plastids:35S::ZmSPK1GFP and 35S::GFP,and transformed them respectively into Arabidopsis by floral dip.Subcellular localization was determined by Confocal laser scanning microscopy.The fused ZmSPK1-GFP showed a nuclear and cytosolic distribution similar to that of free GFP.Compared with 35S::GFP,ZmSPK1GFP was more strongly accumulated in the nucleus,suggesting that ZmSPK1 might be soluble protein and localize in the nucleus.
