Studies on the Virus-free Technology of Stem-tip and Tissue Culture in Allium sativum

Abstract: Virus-free garlics were obtained by cultrring stem-tips of 5 of 5 famous local garlics cultivated in Shanxi. The results showed ththat when the media were MS or B5 supplemented with 1 -2mg/L BA and 0. 01 -0. 1ing/L NAA an,andehof zstem-tip eas 0. 2-0. 5mm, increase of regenerated buds was the most, and that the rratio of the induction of roots was more than 90% when the medium was MS or B5Zcontaining.02mg/L NAA, T. Tsurvival tate was 90% or above after the regenerated plants were transplanted into the energy-saving daylight greenhouse.The results also proved that the virus-free garlic seedlings had not any abnormal changes. They could grow robustly and had a steady heredity and high production through investigating their shapes and examining their chromosomes.
