Changes of Endogenous Hormone in Apple Rootstock During Vitrification in vitro

Abstract: Changes of endogenous hormone in leaves and shoot tips in propagation of Maius pumila were determined by HPLC. The results showed that the contents of GA3, IAA and ABA were very low in vitrified leaves and shoot tips , but CTK contents increased significantly. When plant vitrified heavily CTK contents became very low. During culture period the contents of GA3 and IAA in leaves decresed and CTK contents increased, meanwhile the rate of GA3/CTK,IAA/CTK dropped sharply, about 5 - 10 days later the symptom of vitrication occured. Suppling higher concentration of BA in culture medium caused leaves vitrified and led to GA3 and IAA in leaves dropping. It is concluded that the contents of GA3,IAA and CTK and the rate of GA3/CTK, IAA/CTK was correlated with the Vitrification of apple rootstocks in vitro.
