Roles of AY 9944-A-7 in Inducing in Vitro Maturation of Sheep Oocytes Inhibited by IBMX

Abstract: To evaluate the effects of AY 9944-A-7 on 3-isobutyl-1-methylxanthine (IBMX)-arrested meiotic maturation in ovine oocytes,sheep cumulus oocyte complexes (COCs),split COCs and denuded oocytes (DOs) were cultured in medium supplemented with 250 μmol/L IBMX.Different concentrations of AY 9944-A-7 (0,10,20,30,40 μmol/L) were added into maturation medium respectively to induce meiotic resumption of oocytes.The results showed that,AY 9944-A-7 supplementation in the range from 10 to 40 μmol/L increased the percentage of GVBD and the rate of PBI extrusion significantly in ovine COCs (P<0.01).While AY 9944-A-7 showed no significantly stimulatory effect on the percentage of GVBD and the rate of PBI extrusion of ovine DOs arrested by IBMX (P>0.05).And the highest dose tested (40 μmol/L) resulted in significant degenerations in ovine COCs,split COCs and DOs (P<0.01).The results indicated that AY 9944-A-7 induced meiotic resumption inhibited by IBMX in sheep COCs.But AY 9944-A-7 over dose showed toxic effects on ovine oocytes.The optimal dosage for AY 9944-A-7 to overcome the meiotic inhibition by IBMX and resume meiosis was 20 μmol/L.
