Analysis of SNPs in Promoter Region of Myostatin in Sheep

Abstract: Sequencing along with PCR-RFLP was applied to analyze the polymorphism of MSTN gene promoter?in seven sheep breeds. Two SNPs at -959(T/C)and -784(G/A) including six genotype (TT,TC,CC,GG,GA,?AA)respectively were detected in the promoter region,At -959(T/C),the predominant genotype of TT was identified in Dorset,German Merino and Bashibai sheep,the CC in Bayinbuluke,Duolang and Aletai sheep. A chi-square?analysis suggested that the allele frequencies and genotype frequencies of two polymorphisms were in Hardy-Weinberg equilibrium. Statistical analysis also revealed the variation of intermediate polymorphism information content?(PIC) in six breeds (0. 25 < PIC <0. 50). At -784(G/A),The G was predominant allele in all breeds,whereas?the low polymorphism information content (PIC) was observed in German Meilin and Aletai populations (PIC <?0. 25) and intermediate PIC in the Bayinbuluke,Duolang and Bashibai populations (0. 25 < PIC <0. 50).
