Molecular Detection and Analysis of the Complete Nucleotide Sequence of TYLCV Isolates from Handan and Baoding

Abstract: A new virus disease occurred in Handan and Baoding district in 2009 which caused heavy loss to tomato production.Five samples showing TYLCV-like symptom were collected and the PCR detection result showed that they were tomato yellow leaf curl virus.The DNAβ were not detected from all the samples.Comparison of DNA sequence of Handan and Baoding isolate showed the complete DNA-A sequence of Handan and Baoding were 2 781 bp and shared high sequence identity with other isolates from China between 98.6% and 99.8%.The intergenic region had the highest diversity compared to other regions and shared sequence identity with other isolates from China between 96.2% and 99.7%.Handan isolate was most closely related to Shandong isolate(SD-2),while Baoding isolate was most closely related to the Nanjing isolate(JSNJ-1).Phylogenic analysis based on the DNA-A sequence showed TYLCV isolates from China were classed into two branches and Handan and Baoding isolates belonged to different branches.
