Effects of P1 Infection on the Molecules Associated with Processing and Presentation Function of Porcine Pulmonary Alveolar Macrophages for Exogenous Antigen and Costimulatory Molecules

Abstract: The transcriptional levels of SLA-DR,SLA-DM,CD74 and costimulatory molecules CD40,CD80 and CD86 of porcine pulmonary alveolar macrophages(PAMs) on different phases after porcine circovirus-like agent P1 infection were detected by real-time RT-PCR.The results showed that mRNA levels of all above-mentioned molecules were down-regulated at 3 days post-inoculation(DPI).Among those CD80 mRNA expression showed a downward trend(P0.1),and CD40,CD74 and SLA-DR mRNA expression were significantly declined(P0.05).The mRNA expression of SLA-DR SLA-DM at 8 DPI and SLA-DM at 17 DPI were down-regulated(P0.1).CD74 mRNA expression decreased markedly at 40 DPI(P0.05).Whereas the expression levels of CD86 at 24 DPI showed an upward trend(P0.1).These results suggest that P1 infection affected markedly processing and presentation of PAMs for exogenous antigen,which resulting in a decrease and unbalance of antigen presentation function of PAMs for exogenous antigen and impairment for humoral immune responses.
