cDNA Cloning and Sequence Analysis of Chicken Musclin

Abstract: The coding sequence of chicken Musclin gene was amplified based on the in silico sequence information in the present study. Total RNA was extracted from muscle tissue and mRNA sequence of gene was amplified by RT-PCR using designed primers. The PCR products were ligated into the pMD-19T vector, and then transformed into competent cells of E. coli DH5α. The sequence was analyzed to identify the recombinant plasmid. Identity analysis show that the chicken Musclin nucleotide sequence share 74%, 73% and 70% and the deduced protein sequence share 58%, 54% and 50% homology with that of human, rat and mouse, respectively. The deduced peptide contained a "KKKR" motif and the region homologous to mouse ANP, BNP, CNP. The chicken Musclin gene was successfully cloned in the present study and the sequence has been submitted to GenBank (EF551041).
