Abstract:
The rooting technique on the formation of asparagus tissue culture system were studied in this experiment, including optimization of hormone combination on rooting cultivation, the effects of the new stem subculture, the frequency of transplanting, optimization of hormone combination on subculture of the new stem and the frequency of transplanting have the great effects on rooting regeneration of the following new stem. The best processes of asparagus in vitro are: After induced on J2(MS+NAA 0.2 mg/L+BA 0.5 mg/L), the new stem need subculture one time on J3(MS+NAA 0.2 mg/L+BA 0.1 mg/L) or J9(MS+NAA 0.05 mg/L+BA 0.1 mg/L), Subculture time have the great effect on the following rooting. the new stems which were sub-cultured one time were inoculated on MS+IBA 0.5 mg/L+BA 0.05 mg/L+PP333 2.0 mg/L for rooting differentiation, the rooting rate was obtained 90%. The quantity of roots affects surviving frequency of tissue culture of asparagus. Though optimizing, the root induced rate is obtained 90%-100%, surviving rate 100% after transplanting.
